Is it mandatory to authenticate using kinit explicitly while getting Kerberos conn with Cloudera Hive ODBC driver?

This depends on the AuthMech specified, but if it is set to a mechanism requiring Kerberos auth with HS2s, then the answer is yes.

Manual kinit does work for Kerberos auth with HS2. No issues there.

Let me reframe my question, is there an alternate way to programatically authenticate the connection (similar to UserGroupInformation method in JDBC)?
